过敏试验对环境因素有一定的要求，环境温度缓慢变化，在一定范围内机体可以自行调节而适应。但气温变化过大或过急对豚鼠的健康将产生不良反应，影响过敏试验判断的准确性。在日常试验中，我们会定期监测环境温度，控制在一定范围。本文研究日温差对过敏试验结果的影响。
1 材料与方法
1.1 材料
1.1.1 小牛血清去蛋白注射液（规格：10 mL：0.2 g；批号：20080108；奥鸿药业提供），无菌注射器（购于锦州器化玻站）75%酒精棉球（奥鸿药业提供）、砂轮。
1.1.2 供试用的豚鼠健康合格，体重250～350 g，雌鼠无孕（由辽宁医学院实验动物中心提供）。
1.2 方法
1.2.1 试验分组
选取体重250～350 g健康的豚鼠20小组（每小组6只），分别放在日温差≤4 ℃（对照组）和＞4 ℃（供试组）不同两间饲养室饲养。
1.2.2 环境控制
通过空调控制饲养室温度，其他环境条件两个饲养室保持一致。对照组按要求严格控制温度在18～21 ℃范围内（日温差≤4 ℃），供试组人为调节温度在18～26 ℃ 之间，使日温差＞4 ℃，并通过温度计观察室内温度。
1.2.3 实验方法
用75%酒精棉球擦拭腹腔注射部位，连续3次隔日腹腔注射小牛血清去蛋白注射液0.5 mL，然后每小组选半，分别在第一次注射后第14天及第21天自静脉注射小牛血清去蛋白注射液1.0 mL，注射后30 min内观察不同饲养室两组动物出现过敏反应的情况[1]。
1.3 判定方法
静脉注射供试品30 min内，均不得出现过敏反应，每小组6只中如有竖毛、呼吸困难、喷嚏、干呕连续咳嗽3声等现象中的两种或两种以上者，或有休克、抽搐、死亡现象之一者，应判定为供试品不合格[1]1。
1.4 统计学处理 实验所得数据应用SPSS统计软件进行分析。

动物试验减少了许多人的牺牲，在药学研究过程中，正确选择实验动物、合理的环境因素对实验结果判断是至关重要的。环境因素包括温度、湿度、通风、光照、噪音、笼具、垫料等许多方面。在过敏试验过程中，各种环境因素都有可能影响过敏试验的结果。研究表明，环境温度会影响动物的体温、血糖、基础代谢率等。一旦环境因素变化影响动物造成其机体免疫功能低下时，疾病便可表现出来，在进行动物实验时容易激活一些潜在的病原体引起发病，影响实验结果，造成判断误差[2]。我们在以前的实验中会按国标要求关注控制温度、湿度、气流等环境范围，通过本实验发现，日温差对过敏试验结果也有很大影响。日温差＞4 ℃时，每一个体出现的过敏反应情况明显增多。为了保持实验条件的相对稳定以提高实验的准确性，建议在动物饲养过程中要严格控制日温差。
